Global Study Shows Tagrisso, Orpathys Improve Lung Cancer Survival Significantly

SAFFRON enrolled 338 patients across 230 centers in 29 countries, all with MET overexpression or amplification and disease progression on prior Tagrisso.
In addition to existing China approval via SACHI, the combination has temporary authorization in Switzerland for MET-driven EGFR-mutated NSCLC after progression on EGFR-TKI therapy.

AstraZeneca's Phase III SAFFRON trial showed that combining Tagrisso (osimertinib) with Orpathys (savolitinib) significantly extended both progression-free and overall survival in a hard-to-treat form of lung cancer, Yahoo Finance reported. The trial targeted patients whose tumors kept growing after Tagrisso alone — a common problem driven by a gene change called MET amplification or overexpression.
The results could open the door to global drug approvals. The combination is already cleared in China and has temporary authorization in Switzerland, but SAFFRON's global scope — 338 patients across 230 centers in 29 countries — gives regulators worldwide the data they need, according to ADVFN.
When patients with EGFR-mutated non-small cell lung cancer (NSCLC) stop responding to Tagrisso, one of the most common reasons is MET — a protein that becomes overactive and drives tumor growth. SAFFRON enrolled only patients with this specific MET-driven resistance. That tight focus made the trial's results cleaner and more actionable, according to Directors Talk Interviews.
The trial compared Orpathys 300 mg twice daily plus Tagrisso 80 mg once daily against standard platinum-based doublet chemotherapy. The design was open-label, meaning doctors and patients knew which treatment they received. Both progression-free survival and overall survival improved significantly in the combination arm, ADVFN reported.
Standard chemotherapy requires IV infusions and carries heavy side effects. Tagrisso and Orpathys are both taken as pills. That makes the combination an all-oral regimen — a major convenience for patients. The safety profile seen in SAFFRON was consistent with what the two drugs show individually, Yahoo Finance reported, with no major new concerns flagged.

Switzerland later granted temporary authorization for patients with MET-driven EGFR-mutated NSCLC who progressed on an EGFR inhibitor. SAFFRON now provides the large-scale, multinational evidence that other regulators — including in the US and Europe — typically require, according to Directors Talk Interviews.
The findings reinforce Tagrisso as the backbone of EGFR-mutated lung cancer treatment. They also highlight why MET testing matters. Without knowing whether a patient's tumor has MET changes, doctors cannot identify who will benefit from adding Orpathys. Both AstraZeneca and HUTCHMED stressed the importance of biomarker testing in treatment decisions, Yahoo Finance noted.
Markets responded quickly. HUTCHMED shares rose about 9% intraday after the SAFFRON data were released. The stock traded around HK$20.56, with turnover hitting approximately HK$48.5 million. AstraZeneca also saw positive investor sentiment, reflecting confidence that the trial data could support new revenue from global approvals, according to ADVFN.
For AstraZeneca, SAFFRON adds another potential growth driver to its oncology portfolio. Tagrisso already generates billions in annual sales. Expanding its use into the post-progression MET-driven setting — with a partner drug — could open a meaningful new patient population worldwide, Yahoo Finance reported.
